Driving west to east through the Lowlands

First time in the UK!  We're thinking of driving from Dumfries & Galloway to Edinburgh and/or Dunbar in early July. I've looked at several possible routes via google maps, but I need some expert advice. Can this be realistically done in one day, without traveling the main highways?  Is there a particularly scenic or historical route you would recommend? I haven't seen much info on traveling in the Lowlands. Then there's the question of whether to drive or look for suitable alternate transportation. We would welcome any suggestions!
